| 背景介绍 | Cytokeratin 19 is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21. Immunofluorescence: MCF-7 cells were fixed with 4% paraformaldehyde for 30 min at RT, permeabilized with 0.1% Triton X-100 for 10 min at RT then blocked with 10% goat serum for 30 min at RT. Cells were stained with ARG66324 anti-Cytokeratin 19 antibody [SQab1872] (green) at 1:40,000 and 4°C. DAPI (blue) was used as the nuclear counter stain.

Flow Cytometry: MCF-7 cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% TritonX-100 for 15 min. The cells were stained with ARG66324 anti-Cytokeratin 19 antibody [SQab1872] (red) at 1:200 dilution in 1x PBS/1% BSA for 30 min at RT, followed by Alexa Fluor® 488 labelled secondary antibody. Unlabelled sample (black) was used as a control. .
